一测多评法测定青钱柳中异槲皮苷、槲皮素-3-O-α-L-鼠李糖苷和阿福豆苷的含量 被引量:3

摘要 目的建立超高效液相色谱法测定青钱柳中异槲皮苷、槲皮素-3-O-α-L-鼠李糖苷和阿福豆苷含量的一测多评法。方法以阿福豆苷为内参物,建立阿福豆苷与异槲皮苷、槲皮素-3-O-α-L-鼠李糖苷的相对校正因子,并用该校正因子进行异槲皮苷、槲皮素-3-O-α-L-鼠李糖苷的含量计算(计算值),实现一测多评;同时采用外标法测定药材中3种黄酮含量(实测值),并比较计算值与实测值的差异。结果经测定异槲皮苷、槲皮素-3-O-α-L-鼠李糖苷和阿福豆苷在药材中的含量分别为1.403,1.314,1.633 mg/g;5批青钱柳药材中3种异黄酮的含量计算值与实测值间无统计学意义。结论以外标法测定阿福豆苷,利用相对校正因子实现对异槲皮苷、槲皮素-3-O-α-L-鼠李糖苷含量进行质量评价准确、可行。 Objective To establish UPLC method for the determination of isoquercitrin, quercetin-3-O-α-L-rhamnoside and asafzelin in Cyclocarya paliurus by QAMS. Methods Asafzelin was used as an internal materials, the relative correction factor between asafzelin, isoquercitrin and quercetin-3-O-α-L-rhamnoside was established, And these correction factors were used to calculate the determination of isoquercitrin, quercetin(calculated value) to achieve QAMS; at the same time, the de- termination of 3 kind of flavonoids in herbs (measured value) was measured by the external standard method. Results Through determination, the content of isoquercitrin, quercetin-3-O-α-L-rhamnoside and asafzelin was 1. 403 mg/g, 1. 314 mg/g, 1.633 mg/g respectively; the calculated value of the content of 3 isoflavonoids in 5 batches of cyclocarya paliurus medicinal materials had no significant difference with measured value. Conclusion It was correct and feasible to determinate asafzelin by ESM, and conduct quality evaluation of the content of isoquercitrin and quercetin-3-O-α-L-rhamnoside by using the relative correction factors.
