法舒地尔在动脉粥样硬化中对超氧化物歧化酶的影响

摘要 目的探讨法舒地尔在大鼠动脉粥样硬化形成过程中对超氧化物歧化酶(SOD)及丙二醛(MDA)的影响。方法将40只健康雄性Wistar大鼠(体质量220~250 g)分为正常组、动脉硬化组和法舒地尔组和阿托伐他汀组,各10只。正常组给予正常饮食;余3组每只大鼠均给予维生素D3+高脂饮食;法舒地尔组同时给予法舒地尔5 mg/kg体质量,每天2次腹腔注射;阿托伐他汀组在上述饮食基础上同时给予阿托伐他汀片5 mg·kg^-1·d^-1喂养。9周后空腹24 h抽血并处死,检测血脂水平,自主动脉弓下约1 cm处留取动脉组织1 cm用4%甲醛溶液固定,行HE染色。结果动脉硬化组均形成了典型的粥样斑块;但法舒地尔组及阿托伐他汀组中动脉粥样硬化明显减轻,且血清MDA含量明显减少(均P〈0.01),SOD含量显著增加(均P〈0.01)。结论法舒地尔的抗动脉粥样硬化作用可能与增加SOD的活力,减少MDA含量,抑制脂质过氧化过程对血管内皮的损害,从而延缓和阻止AS斑块的形成和发展。
