摘要
为建立小麦(Triticum aestivum Linn.)幼胚的转化体系,获得转基因的小麦再生植株,对小麦幼胚进行愈伤组织诱导。以新春6号和新春9号小麦幼胚为试验材料,选择不同诱导培养基对幼胚进行愈伤组织诱导。结果表明,4种培养基诱导的愈伤组织质量和诱导率有明显的差异,其中MCIMA+KT高于其他3种培养基,新春6号和新春9号诱导率分别为53.80%和52.25%,愈伤组织的质量级别为A级,属于极适宜进行遗传转化状态。SD2培养基上诱导出的2个小麦品种愈伤组织分别为0.002%和0.59%,是不适宜进行遗传转化的培养基。新春6号和新春9号2个基因型诱导的愈伤组织数量和质量之间没有明显差异,适于进行遗传转化。
To estabhsh the transformation system of wheat immature embryos, and regenerate transgenic wheat plants, Xinchun 6 and Xinchun 9 immature embryos of wheat were used to screen medium for callus induction of immature embryos with different induction. The results showed that 4 kinds of culture media had significant differences between quality and in- duction rate of the callus induced. MCIMA + KT was significantly higher than other three with induction rate of 53.80% and 52.25% and the A level of callus. It was suitable for genetic transformation. The induction rate of the SD2 callus was 0.002% and 0.59% and was not appropriate for genetic transformation of the culture medium. Xin Chun 6 and XinChun 9 wheat had no obvious difference ,between quantity and quality on two genotypes of callus induced, both were suitable for genetic trans- formation. The best suitable time for immature embryos culture of spring wheat Xinchun 6 and XinChun 9 were 3-4 d after flowering.
