摘要
采用PDA培养基、PDA-抗生素培养基以及MMN培养基,通过切片法从一年生樱桃(Prunus pseudocerasus)根中分离菌根真菌,用打孔法分离得到纯化菌株,并对菌株的生理生化特征进行系统鉴定。结果表明,切片分离时的表面消毒条件和方法以及分离培养基的选用均直接影响菌根真菌的分离纯化结果及菌根真菌的多样性。该菌利用HgCl处理6 min,接种在改良MMN培养基中分离效果最好,该菌的最适pH 7,最适温度为25℃,最适P浓度KH2PO4和K2HPO4分别为0.50 g/L和1.00 g/L,最适C源为可溶性淀粉,最适N源为尿素。将该菌加入到生长基质中能将炼苗期的樱桃苗的成活率由73%提高到86%。
The process of isolating micorrhizal fungi from cherry root by sectioning was evaluated and the isolated fungi were identified. The isolated fungi were cultivated in the PDA medium, PDA-antibiotic medium and MMN medium and subsequently purified by punch method. The results showed that disinfection conditions and methods of slice surface and medium selection affected directly the purification and diversity of the fungi isolated. The best isolation was on the modified MMN medium as isolated fungi were sterilized by HgC1 for 6 minutes. The isolated fungi could better growth at 25 ℃, pH 7.0, KH2PO4 0.50 g/L and K2HPO4 1.00 g/L. Soluble starch and urea were the best sources of carbon and nitride. Survival rate of cherry seedlings increased from 73 % to 86% after adding the isolated fungi into sub- strate.
