酸性成纤维细胞生长因子的分离纯化及活性分析

摘要 以瞬时表达酸性成纤维细胞生长因子(aFGF)的豌豆(Pisum sativum L.)为试验材料,提取豌豆中可溶性总蛋白质,利用Western blot方法检测目的蛋白质,结果表明豌豆可成功表达aFGF;采用肝素亲和柱层析法分离目的蛋白aFGF,并用MTT法检测aFGF的生物学活性,结果表明豌豆中表达的酸性成纤维细胞生长因子具有促细胞分裂活性。 The pea(Pisum sativum L.) plants expressing the foreign protein aFGF were used to extract the total soluble pro- teins. Western blot was performed to identified the aFGF protein. Heparin-affinity chromatography was used to separate and purify the aFGF. The bioactivity of the aFGF purified was analyzed by MTT. The result of western blot confirmed that the exogenous protein aFGF was expressed and purified successfuly from pea plants. The MTT results showed that the pea plants derived aFGF had the mitogentic activity.
