摘要
通过筛选的10条引物对供试的18种水稻材料进行PCR扩增。共扩增出清晰、重复性好的谱带144个,其中多态性带为138条,多态性比率高达95.8%;应用引物808、809、810、816及其组合获得了每个品种的ISSR特征带,有效区分出所有供试材料,并建立18个水稻品种的ISSR指纹图谱;统计10条ISSR引物的标记信息,对18个水稻样品的遗传相似性进行计算,得出遗传相似性系数在0.42-0.94之间存在显著的遗传变异,平均相似性系数为0.680。通过UPGMA法构建的树状聚类图,在相似性系数为0.719时可以将18个供试样本以粳、籼稻严格地划分为2个大类群。
Through 10 screened primers selected 18 species of rice material for PCR amplification. 144 bands with distinct and good repeat ability were amplified, which was 138 polymorphic bands and the polymorphic rate was up to 95.8%. Applications and combinations of primers 808,809,810, 816 and their combination won ISSR features with each species could effectively distinguish all the tested materials and establish their ISSR finger-print. Statistics 10 ISSR primers tag information and calculating the 18 samples of rice genetic similarity, the clustering analysis showed that rice samples tested genetic similarity coefficient of 0.42 to 0.94 had a significant genetic variation, with an average similarity coefficient was 0.680. The 18 tested rice cultivars could be strictly divided into two types of Indica and Japonica when the similar coefficient was 0.719 in tree dendrogram constructed by UPGMA method.
