摘要
通过正交试验和中心复合设计试验优化了水浴法和超声辅助法提取连钱草总黄酮的提取工艺,并对两种方法进行比较.结果表明,水浴法提取的最优工艺是:液固比20∶1(mL/g)、时间90 min、温度40℃、乙醇浓度85%;超声辅助法提取的最优工艺是:液固比23∶1(mL/g)、超声功率170 W、超声时间11 min、乙醇浓度85%.对两种方法的比较可以得出:超声辅助提取的平均得率为(8.63±0.25)%,而水浴法提取的平均得率为(6.07±0.12)%,黄酮提取率提高了42.17%,因此,超声辅助提取连钱草总黄酮产量高于传统的水浴法,而且还具有节能、省时的优点,该研究为连钱草总黄酮提取的工业化生产提供了理论基础.
In present study, the processes of extracting total flavonoids from Glechomae by water bath and ultrasonic-assisted extraction were optimized using orthogonal test and central composite design experi-ment, respectively.Meanwhile, the two methods were compared.The results showed that the optimal process of bath extraction was as follows: liquid-solid ratio 20 ∶1(mL/g), time 90 min, temperature 40℃and ethanol concentration 85%.Meanwhile the optimal technology parameters of ultrasonic-assisted extraction were liquid-solid ratio 23 ∶1( mL/g) , ultrasonic power 170W , time 11 min,and ethanol con -centration85%.The yield of total flavonoids from Glechomae by ultrasonic-assisted and water bath meth-ods was(8.63±0.25)%and (6.07±0.12)%,respectively,and the yield of ultrasonic-assisted method was elevated by 42.17%compared to that of bath extraction.In conclusion, the ultrasonic-assisted extraction of total flavonoids from Glechomae not only can improve the yield but also save energy and time.The present study provides the theoretical basis for the industrial production of Glechomae total flavonoids.
