胡椒酸合成工艺研究

摘要 以胡椒碱为原料,在碱-乙醇溶液中加热回流制备胡椒酸酯类降血脂药物的重要中间体胡椒酸,探讨了反应时间、碱的种类、料液比以及碱浓度对胡椒酸产率的影响,确定较佳的合成工艺条件.制备胡椒酸的最佳合成工艺条件为:选择氢氧化钾作为催化剂,氢氧化钾乙醇溶液的浓度为15%,料液比为1∶1.2,反应时间为5 h.通过对合成产品的熔点和红外光谱分析,确定所得产品为胡椒酸,纯度可达95%以上,产率可达到90%. Taking piperine as raw material, we prepared an important intermediate of hypolipidemics of piperic acid esters, i.e. piperic acid, in an alkali-ethanol solution by refluxing under heating, and discussed the effects of reaction time, the kind of alkali, material-to-liquid ratio, and alkali concentration on the yield of piperic acid to determine the optimum synthesis conditions. The results showed that the optimum synthesis conditions were as follows: catalyst KOH, KOH-ethanol solution concentration 15%, material-to-liquid ratio 1:1.2, and reaction time 5 hours. Through melting point determination and IR spectroscopy analysis, we determined that the synthesized product was piperic acid; the purity was above 95%; and the yield reached 90%.
