摘要
目的探讨神经母细胞瘤细胞(SH-SY5Y)和皮层神经元对神经毒物乙酸铅和硫酸铝敏感性的差异,为神经元体外替代方法及实验研究提供一定的数据支持。方法原代培养皮层神经元及培养细胞系SH-SY5Y,乙酸铅和硫酸铝染毒浓度分别为25、50、100、200、400、800、1600、3200μmol/L,作用时间分别为3和24h,测定并计算细胞存活率和半数致死浓度(IC50)值。结果除硫酸铝作用24h外,皮层神经元的IC50值均低于SH-SY5Y;SH-SY5Y对乙酸铅IC50值均高于硫酸铝;皮层神经元对乙酸铅的IC50值低于硫酸铝。结论皮层神经元对乙酸铅和硫酸铝的毒性较SH-SY5Y敏感。若以皮层神经元为靶细胞,则乙酸铅相对敏感,若以SH-SY5Y作为靶细胞,则硫酸铝则更为敏感。
Objectives To explore the differences of sensitivity of neuroblastoma (SH-SY5Y) and cortical neurons to neurotoxicity chemicals (lead acetate and aluminium sulfate) and provide data support for alternative methods of neurons in vitro and experimental study. Methods Primary culture cortical neurons and cell line SH-SY5Y were used and treated with lead acetate and aluminium sulfate at the doses of 25, 50, 100, 200, 400, 800, 1 600 and 3 200 μmol/L for 3 h and 24 h respectively, the cell viability and IC50 were calculated. Results IC50 value of aluminium sulfate for cortical neurons was less than that for SH- SY5Y, and as for lead acetate, it was higher than that for SH-SY5Y; IC50 value of lead acetate was lower than that of aluminium sulfate for cortical neurons. Conclusion Cortical neurons are more sensitive to two chemicals. As the tested ceils, cortical neurons are more sensitive to lead acetate, SH-SY5Y cells are more sensitive to aluminium sulfate relatively.
