重组人Ⅱ型肿瘤坏死因子受体-抗体融合蛋白对胶原诱导性关节炎大鼠模型的抗炎作用 被引量:2

Study on the anti-inflammatory action of recombinant human tumor necrosis factor-α receptor Ⅱ∶Fc in CIA model rats

摘要 目的:探讨重组人Ⅱ型肿瘤坏死因子受体-抗体融合蛋白(rhTNFR∶Fc)对牛Ⅱ型胶原诱导性关节炎(CIA)大鼠关节炎的抗炎作用。方法:将40只6周龄雌性Wistar大鼠随机分为空白对照组、模型对照组、治疗组和醋酸泼尼松对照组,每组10只。制备CIA模型,观察rhTNFR∶Fc对大鼠足趾肿胀度、关节炎指数变化及血清TNF-α、IL-1β、IL-10、IL-17的水平的影响。结果:rhTNFR∶Fc能够减轻CIA大鼠足趾肿胀,降低关节炎指数,并能降低血清TNF-α、IL-1β及IL-17水平,上调IL-10的水平,和醋酸泼尼松对照组相比,无明显差异(P<0.05)。结论:rhTNFR∶Fc对CIA大鼠关节炎具有明显的抗炎作用,其机制可能与其下调TNF-α、IL-1β及IL-17水平,上调IL-10的水平有关。 OBJECTIVE To study the anti-inflammatory action of recombinant human tumor necrosis factor-α receptorⅡ∶ Fc(rhTNFR∶ Fc) in CIA(collagen induced arthritis) model rats.METHODS 40 Six-week-old female Wistar rats were divided randomly into 4 groups:blank control group(n = 10),model control group(n = 10),treatment group with rhTNFR:Fc(n = 10) and prednisone acetate control group(n = 10).RESULTS Through establishing CIA model,and observing the effect of rhTNFR∶ Fc on the change of rats toes' swollen degree,arthritis index and expression level of TNF-α,IL-1β,IL-10 and IL-17 in serum,it was discovered that rhTNFR:Fc could alleviate swollen degree of CIA rats toes,lower index of arthritis,reduce expression level of TNF-α,IL-1βand IL-17 and increase expression level of IL-10 in serum.Compared with prednisone acetate control group,there was no visible difference(P〉0.05).CONCLUSION There is a visible anti-inflammatory action of rhTNFR:Fc on CIA model rats,the mechanism of which may relate to reduce expression level of TNF-α,IL-1β and IL-17 and increase expression level of IL-10 in serum.
