补肾平喘膏方对支气管哮喘SD大鼠Th17相关细胞因子的调节作用

Regulating Action of Bushen Pingchuan Cream Formula for the Related Cytokine of Th17 in Rats with Bronchial Asthma

目的:观察支气管哮喘(简称哮喘)大鼠血浆中Th17相关细胞因子IL-6、IL-17、TGF-β水平及补肾平喘膏方干预后的变化情况,以探讨补肾平喘膏方防治哮喘的作用机制。方法:60只SD大鼠随机分为正常对照组、模型组、地塞米松组、膏方预防组、膏方治疗组和膏方防治(预防加治疗)组,每组10只。除正常对照组外,各组大鼠以卵白蛋白致敏并诱发哮喘模型。正常对照组与模型组生理盐水灌胃,其他各组采用药物干预,Elisa夹心法测定各组大鼠血浆中IL-6、IL-17、TGF-β的表达。结果:模型组大鼠血浆中IL-6、IL-17和TGF-β的水平高于正常对照组(P<0.01)。地塞米松组及膏方各组IL-6、IL-17和TGF-β的水平较模型组降低(P<0.05)。膏方各组间无明显统计学差异(P>0.05)。膏方各组下降明显,与地塞米松组对比具有统计学差异(P<0.05)。结论:补肾平喘膏防治哮喘的作用机制,可能与其调节哮喘大鼠血浆中Th17相关细胞因子的作用有关。

Objective： To observe the effects of Bushen Pingchuan Cream Formula on related cytokine of Thl7 in rats＇ plasma with asthma, and to explore its mechanism in treating asthma. Methods： Sixty SD rats were randomly divided into normal controlled group, untreated group, dexame thasone group, and medicinal extract prevented, medicinal extract treated, and medicinal extract prevented and treated groups, with ten rats in each group. Asthma was induced by intraperitoneal injection of ovalbumin （OVA） and forced inhalation of atomized OVA. Expression of IL-6, IL-17 and TGF-β in blood was detected by Elisa method. Results： Expression of IL-6, IL-17 and TGF-β in blood was higher in the untreated group than in the normal controlled group （P 〈0.01 ）. Expressions oflL-6, IL-17 and TGF-β in medicinal extract groups were down regulated as compared with those in the untreated group and dexamethasone group （P 〈0. 05）, and there were no significant differences as compared with the normal controlled group （P 〉0. 05）. Conclusion： Bushen Pingchuan Cream Formula can control and prevent the transformation of Thl7, which may be one of its mechanisms in preventing and treating asthma.
