摘要
目的:建立测定人血清中顺式阿曲库铵浓度的方法。方法:血清酸化后采用二氯甲烷液-液萃取法处理,以高效液相色谱(HPLC)-荧光法进样测定,色谱柱为Kromasil100-5C18,流动相为1.0%三氟乙酸-乙腈(58∶42),流速为0.8ml/min,柱温为常温,荧光检测波长分别为280 nm(E x)、320 nm(E m)。结果:顺式阿曲库铵血药浓度在15.625~2 000ng/ml范围内线性关系良好(r=0.999 3),平均提取回收率为78.78%~83.59%,平均方法回收率为98.58%~101.23%,日内、日间RSD均小于10%。结论:本方法操作简便、灵敏度高、重现性好、结果可靠,可满足临床上血清中顺式阿曲库铵血药浓度检测和药动学研究的需要。
OBJECTIVE: To establish a method for the determination of cisatrcurium concentration in human serum. METHODS: The serum sample was extracted by dichloromethane liquid-liquid extraction after acidification. HPLC-flurorescence method was adopted. The determination was performed on Kromasil 100-5 C18 column with mobile phase consisted of 1.0% TFA-acetonitrile (58:42) at the flow rate of 0.8 ml/min. The column temperature was normal atmospheric temperature; the detection wavelength was set at 280 nm (Ex) and 320 nm (Em). RESULTS: The linear range of cisatracurium was 15.625-2 000 ng/ml (r=0.999 3). Av- erage extraction recovery was 78.78%-83.59%, and average method recovery was 98.58%-101.23%, respectively. The inter-day and intra-day RSDs were less than 10%. CONCLUSIONS: The method is simple, sensitive, reproducible and reliable, and can meet the demand of concentration determination and pharmacokinetics study of cisatracurium.
