摘要
目的:观察茶多酚(TP)在脂多糖(LPS)介导下对人牙周膜成纤维细胞(HPDLCs)MMP-1、MMP-2表达的影响。方法:体外培养HPDLCs,在培养液中添加TP(200μg/ml)作用于脂多糖(100μg/ml)介导的HPDLCs,培养24、48、72 h,ELISA法检测HPDLCs分泌MMP-1和MMP-2的量。荧光实时定量PCR法检测HPDLCs中MMP-1和MMP-2 mRNA的表达。结果:在LPS介导下HPDLCs MMP-1和MMP-2分泌量和基因表达升高,TP可抑制LPS介导下HPDLCs MMP-1和MMP-2表达。结论:TP可抑制LPS介导的人牙周膜细胞的胶原降解。
Objective: To survey the expression of MMP-1,MMP-2 of human periodontal ligament cells(HPDLCs) treated by tea polyphenols(TP) and lipopolysaccharide(LPS). Methods: HPDLCs were in vitro cultured in vitro and treated by TP(200 μg / ml)and / or LPS(100 μg / ml) for 24,48 and 72 h respectively,the secretion of MMP-1 and MMP-2 were examined by ELISA,MMP-1and MMP-2 mRNA expression was examined by real-time PCR. Results: The secression and mRNA expression of MMP-1 and MMP-2 of HPDLCs increased by LPS treatment and significantly inhibited by TP at the different times. Conclusion: TP can inhibit the collagen degradation of HPDLCs mediated by LPS.
