风湿性心脏病心房颤动患者心房肌HCN2通道的表达变化

The expression of atrial hyperpolarization-activated cyclic nucleotide-gated channel 2 in atrial fibrillation patients with rheumatic heart disease

摘要 目的通过研究风湿性心脏瓣膜病(风心病)心房颤动(Af)患者心房肌超极化激活环核苷酸门控通道-2(HCN2)的表达变化,探讨其与Af发生维持的关系,为分析Af发生维持的机制奠定理论基础。方法选取风心病二尖瓣狭窄患者42例,根据是否合并Af将其分为两组,Af组23例,窦性心律组19例,术中取右心耳组织,应用蛋白质免疫印迹法(Western blot)分别测定两组HCN2通道的表达水平。结果 Af组和窦性心律组中HCN2通道在心房肌组织中的相对表达量分别是(0.45±0.19)和(0.24±0.12),两组间比较,差异有统计学意义(P<0.05)。结论心房肌组织中HCN2通道的表达上调,可能是风心病Af的分子机制之一,参与调控Af的发生和维持。 【Objective】To explore the mechanism of atrial fibrillation through evaluating the expression of HCN2 channelin muscles of right atrium in rheumatic heart disease patients with atrial fibrillation. 【Methods】Right atrial appendages were obtained underwent cardiac valve replacement surgery in two groups of rheumatic heart disease patients with mitral valve stenosis, one group with atrial fibrillation and another group with sinus rhythm, and the atrial expression of HCN2 channelwas measured by Western bolt. 【Results】 The relative expression of HCN2 channelin experimental and control groups was(0.45±0.19) and(0.24±0.12), and the difference was statistically significant(P〈0.05).【Conclusion】The overexpression of HCN2 channel might participate in the progress of atrial fibrillation development in rheumatic heart disease, which reveals the molecular mechanism.
