A method of extracting total RNA from iliac cancellous bone with scoliosis

为了创建一种高效提取人体骨组织总RNA技术,分别采用Trizol、RNeasy lipid kit及改良CTAB提取人体松质骨总RNA,并分别用核酸蛋白检测仪、1%琼脂糖凝胶电泳和荧光Real-time PCR检测3种不同方法提取的总RNA质量。结果显示,3种方法中,Trizol方法提取的总RNA纯度最差,RNeasy试剂盒提取的总RNA纯度得到提高,但完整性差,改良的CTAB方法提取的总RNA纯度和完整性符合分子生物学实验要求。改良的CTAB法是从骨组织中提取总RNA高效便捷的方法,适于临床骨组织基因表达的研究。

To establish an efficient method for extraction of total RNA from human bone tissue ,total bone RNA with 3 different extraction methods were detected by Biophotometer plus .1% agarose gel electrophoresis and fluorescent Real-time PCR .The results show that the total RNA purity is the worst with Trizol method ,the integrity is the worst and the purity is improved with RNeasy kits method , the purity and integrity with modified CTAB methods are both satisfied with the requirements of the molecular biology experiment .The modified CTAB method of total RNA isolation is efficient and convenient ,which is suitable for gene expression studies in clinical samples of bone tissue .
