趋化因子受体CXCR4、PTEN在结肠黏液腺癌中的表达及临床意义

摘要 目的探讨趋化因子受体CXCR4和PTEN蛋白在结肠黏液腺癌中的表达及其与癌转移及预后的关系。方法采用免疫组织化学及组织芯片技术,检测25例结肠正常组织、92例大肠黏液腺癌组织及癌旁组织中PTEN蛋白和趋化因子受体CXCR4的表达情况,并与临床资料对比分析。结果 PTEN蛋白在结肠黏液腺癌的表达率为31.52%(29/92),在癌组织和癌旁组织的表达率明显低于正常结肠组织,差异有统计学意义(P<0.05);趋化因子受体CXCR4在结肠黏液腺癌的表达率为61.95%(57/92),在癌组织和癌旁组织的表达率明显高于正常结肠组织,差异有统计学意义(P<0.01)。PTEN蛋白、CXCR4与肿瘤浸润深度、淋巴结转移和临床分期显著相关(P<0.05)。结论趋化因子受体CXCR4和PTEN蛋白的过表达与结肠癌的浸润、转移相关,且它们在其浸润转移中具有相关性,常态化检测有利与结肠黏液腺癌的治疗。 Objective To investigate the expression and clinical significance of chemokine receptor-CXCR4,PTEN in colon mucinous adenocarcinoma and to explore the correlation between their expression and cancer invasion,metastasis as well as patient’ s prognosis.Methods The expression levels of PTEN protein and CXCR4 were detected by immunohistochemistry in 20 cases of normal colon tissues, 92 cases of colorectal mucinous adenocarcinoma and the tissues beside cancer.The detection results were contrastively analyzed with clinical data.Results The positive expression rate of PTEN protein in colon mucinous adenocarcinoma was 31.52%(29/92),which in cancer tissue and the tissues beside cancer was significantly lower than that in normal colon tissue ( P〈0.05).The positive expression rate of CXCR4 in colon mucinous adenocarcinoma was 61.95%(57/92),which in cancer tissue and the tissues beside cancer was significantly higher than that in normal colon tissue ( P 〈0.01) .The expressions of PTEN protein and CXCR4 were closely correlated to the infiltration depth of cancer,lymph node metastasis and tumor staging( P 〈0.05).C onclusion The overexpression of chemokine receptor CXCR4 and PTEN protein are closely related to invasion,metastasis of colon carcinoma,so routine detection of chemokine receptor CXCR4 and PTEN protein is helpful for the treatment of colon mucinous adenocarcinoma.
