摘要
目的探讨红花注射液对脑缺血再灌注损伤大鼠大脑皮质Bcl-2及Bax蛋白表达的影响。方法选取36只健康雄性SD大鼠,随机分为假手术组,模型组,红花注射液低剂量组[(0.8 mL/(kg·d)]、中剂量组[1.6 mL/(kg·d)]、高剂量组[(3.2 mL/(kg·d)]及尼莫地平注射液组[2.0 mL/(kg·d)],每组各6只。假手术组及模型组均给予等体积生理盐水。各组大鼠均于术前1周每天上午9:00灌胃给药1次,连续7 d。采用线栓法建立大鼠局灶性脑缺血再灌注损伤模型;采用免疫组织化学方法检测大鼠脑皮质Bcl-2及Bax蛋白的表达。结果假手术组未见脑皮质Bcl-2、Bax阳性细胞表达,模型组Bcl-2、Bax阳性细胞表达明显,两组阳性细胞数比较,差异有统计学意义(P<0.05);与模型组比较,红花注射液低、中、高剂量组和尼莫地平注射液组Bcl-2阳性细胞表达明显增多,且红花注射液高剂量组增多最明显,差异有统计学意义(P<0.05),而Bax阳性细胞表达均明显降低,差异有统计学意义(P<0.05),但红花注射液低、中、高剂量组间比较,差异无统计学意义(P>0.05)。结论红花注射液可通过增加Bcl-2和下调Bax表达减少大脑神经细胞凋亡,对缺血再灌注损伤大鼠脑组织发挥保护作用。
Objective To explore the influence of safflower injection on expression of Bcl-2 and Bax protein in the cor-tex of rats after ischemia-reperfusion injury. Methods Thirty-six healthy male SD rats were selected and randomly divided into sham-operation group,model group,low-dose group of safflower injection[(0.8 mL/(kg&#183;d)],middle-dose group[1.6 mL/(kg&#183;d)], high-dose group [3.2 mL/(kg&#183;d)] and nimodipine injection group[(2.0 mL/(kg&#183;d)],6 cases in each group. The sham-operation group and model group were managed with equal volume of normal saline instead. All the groups administrated with gastric injec-tion at 09:00 1 week before operation for 7 d continuously,once daily. To establish the focal cerebral ischemia-reperfusion injury model by thread technique;the expressions of Bcl-2 and Bax protein in the brain cortex of rats were detected by immunohisto-chemical methods. Results The positive expression of both Bcl-2 and Bax protein were significantly increased in the model group,compared with the negative result in the control group,the difference had statistical significance(P〈0.05). Compared with the model group,the positive expression of Bcl-s in low-dose group,middle-dose group and high-dose group of safflower injection and nimodipine injection group increased obviously,the increase of the high-dose group of safflower injection was the most obvi-ous,and the difference was statistically significant (P〈0.05),but the difference among low-dose group,middle-dose group and high-dose group had no statistical significance (P〉0.05). Conclusion Safflower injection has protective effect on brain tissue of rats after ischemia-reperfusion injury by up-regulating Bcl-2 and down-regulating Bax to reduce apoptosis of cerebral cortical neurons.
