大鼠脑出血后脑红蛋白在脑组织中的表达及其对细胞凋亡的影响 被引量:2

摘要 目的研究实验性大鼠脑出血后脑组织中脑红蛋白的表达及其对细胞凋亡的影响。方法随机将86只大鼠分为正常对照组、假手术组及脑出血组,应用立体定向技术,将自体未抗凝血注入大鼠基底节区制备脑出血模型。分别在造模后6、24、48、72小时断头取脑制作标本,采用TUNEL染色检测脑组织中细胞凋亡、荧光定量RT-PCR检测脑红蛋白mRNA表达、免疫组化分析脑红蛋白的表达。结果脑出血后6h血肿周围脑组织脑红蛋白及TUNEL染色阳性细胞表达升高,其中脑红蛋白48h达高峰,72h开始下降,而细胞凋亡72h达高峰,表达均明显高于假手术组和正常对照组,差异有统计学意义(P<0.05,<0.01);脑红蛋白的表达与细胞凋亡数呈正相关(r=0.51,P<0.05。结论脑出血后脑红蛋白表达上调,可能对脑出血继发神经损伤起重要的保护作用。 Objective To study the expression of neuroglobin in brain tissue of rats with experimental intracerebral hemorrhage and its effect on apoptosis.Methods 86 rats were randomly divided into control group,sham-operated group and intracerebral hemorrhage group.Intracerebral hemorrhage was induced by stereotaxic injection of autologous blood into right caudate nucleus of the rats; Rats were killed at 6,24,48 and 72 hours after the establishment of animal models respectively.TUNEL method was used to detect apoptosis,while both RT-PCR and immuno-histochemistry were used to detect the expression of neuroglobin in brain tissues at different times after intracerebral hemorrhage.Results The expression of neuroglobin and TUNEL-positive cells in peri-hematomal brain tissue started to increase at 6 hours,the expression of neuroglobin peaked at 48h,and began to decrease at 72h.But,apoptosis peaked at 72h.which were all obviously higher than those in the sham-operated group and normal control group,neuroglobin expression was positively correlated with apoptosis cells.Conclusions The expression of neuroglobin in brain tissues was up-regulated after intracerebral hemorrhage,and it participated in the pathophysiologic course of intracerebral hemorrhage.It played an important protective effect against secondary neuronal injury.
