半胱氨酸对玻璃化卵子冷冻损伤的改善作用

摘要 目的探讨半胱氨酸对玻璃化卵子冷冻后发育潜能及纺锤体正常形态率的研究。方法在玻璃化冷冻卵子中添加1 mmol/L半胱氨酸,观察其发育潜能。同时利用纺锤体观察仪计算纺锤体正常形态率。最后选取部分卵子固定染色后,经激光共聚焦显微镜观察其形态变化。结果 1 mmol/L半胱氨酸能有效提高玻璃化卵子冷冻后受精率(70.8%和84.8%)(P<0.05),但对3-4细胞和桑葚+囊胚获得率无明显改善作用(P>0.05)。同时半胱氨酸提高了复苏后卵子纺锤体正常形态率(53.3%和74.4%)(P<0.05)。结论玻璃化冷冻引起卵子发育潜能下降,异常纺锤体增加。半胱氨酸可以有效提高玻璃化冷冻卵子受精率及纺锤体正常形态率。 Objective: To study the effect of cysteine on developmental potential and spindle morphology of vitrified oocytes. Methods: Oocytes were vitrified with 1 mmol/L cysteine or without cysteine before vitrification, compared with fresh oocyte. The developmental potential was observed. Meanwhile, spindle morphology was analysed by polscope and detected by laser scanning microscopy. Results: lmmol/L cysteine can effectively improve the fertilization rate of vitrified oocytes (70.8% VS 84.8%) (P〈0.05) , which cannot increase 3-4 cell rates and morula +blastocysts rates (P〉0.05) . There were also significantly increased in normal spindle morphology rate (53.3% VS 74.4%) (P〈0.05) . Conclusion: Vitrification caused developmental potential of oocytes to drop and abnormal spindle to increase. Cysteine can effectively improve fertilization rate and normal spindle morphology rate of vitrified oocytes.
