以滤纸酶活力为指标优化解淀粉芽孢杆菌Tu-115菌株产纤维素酶液体发酵条件 被引量:9

摘要 以滤纸酶活力为指标对解淀粉芽孢杆菌(Bacillus amyloliquefaciens)Tu-115菌株产纤维素酶液体发酵条件进行优化。通过单因素和正交试验相结合的方法,考察培养基中碳源、氮源、起始pH值、装瓶量、温度、接种量、发酵时间等发酵条件对产酶能力的影响。结果表明,当培养条件为葡萄糖4%、黄豆饼粉3%、培养基起始pH值4.0、接种量4%、装瓶量50 ml/250 ml,37℃发酵72 h时,发酵液中滤纸酶活力达到最高值,为3 309.90 U/ml,比优化前提高了125倍。优化后,Tu-115菌株产滤纸酶能力大大提高,达到了预期目的。 With filter paper enzyme as index, op- timization of liquid state fermentation condition for producing cellulase by Bacillus amyloliquefa- ciens strain Tu-ll5 was done. Fermentation con- ditions, such as carbon source, nitrogen source, medium initial pH value, media volume, tempera-ture, inoculation volume, fermentation time, which could affect cellulase production by strain Tu-115, were optimized by the method of single factor experiment and orthogonal test. Results showed that cel- lulase quantity reached 3 309.90 U/ml when the glucose was 4%, yellow bean cake powder was 3%, medium initial pH was 4.0, inoculation volume was 4%, media volume was 50 ml/250 ml, fermentation for 72 h at 37 ℃. With the optimization of medium for fermentation, cellulase quantity was 125 times higher than before, achieving the desired aim.
