痰热清注射液对Lewis肺癌化疗模型小鼠免疫功能的影响

The effects of Tanreqing-injection on immune function of Lewis lung carcinoma mouse model treated with chemotherapy

目的研究痰热清注射液对Lewis肺癌化疗模型小鼠免疫功能的影响探讨该药对肺癌机体的免疫调节作用。方法构建小鼠Lewis肺癌模型,分为模型对照组、单独化疗组、联合痰热清化疗组及正常对照组。每组均为8只小鼠。各组治疗后,摘眼球取血、脾脏和胸腺观察各组小鼠脾脏和胸腺指数及外周血淋巴细胞计数、外周血淋巴细胞亚群比例、脾淋巴细胞增殖活性、外周血血清中IFN-γ、TNF-α、IL-4的水平及小鼠脾NK细胞杀伤活性。结果化疗后,联合痰热清化疗组小鼠脾指数、胸腺指数及外周血淋巴细胞计数明显高于单独化疗组(P<0.05);联合痰热清组小鼠外周血CD4+T细胞比例明显高于单独化疗组而CD4+CD25+T细胞比例明显低于单独化疗组(P<0.01);痰热清组小鼠外周血IFN-γ、TNF-α含量明显高于单独化疗组(P<0.01),同时脾淋巴细胞增殖活性和NK细胞杀伤活性明显高于单独化疗组(P<0.01)。结论痰热清注射液对肺癌化疗机体的免疫系统有明显保护作用,并对抗肿瘤免疫力有明显的促进作用。

To investigate the effect of Tanreqing-injection on immune function of mouse model of lung carcinoma treated by chemotherapy and discusse the immunoregulation function of the herb,mouse model of Lewis lung carcinoma was built,and mice were divided into the normal control group,the model control group,the chemotherapy group and the chemotherapy combined with Tanreqing-injection group,which were treated by corresponding strategy.Then the mice of all groups were sacrificed,and the peripheral blood,spleen and thymus were separated to detect peripheral blood lymphocytes count,spleen index（SI）,and thymus index（TI）.Furthermore,the proliferation of spleen lymphocytes and the cytotoxicity of NK were investigated by MTT method;the subsets of peripheral blood lymphocytes were detected by flow cytometry（FCM）;the levels of IFN-γ,TNF-α,IL-4 in blood serum were measured by ELISA.Results showed the SI,TI and lymphocyte count of chemotherapy combined with Tanreqing-injection group were higher than those of the chemotherapy group obviously（P〈0.05）.And the chemotherapy combined with Tanreqing-injection group demonstrated notable improvement in peripheral-blood lymphocytes subset,proliferation of spleen lymphocytes and cytotoxicity of NK,as compared with the chemotherapy group（P〈0.01）.Meanwhile,the levels of IFN-γ and TNF-α in blood serum of chemotherapy combined with Tanreqing-injection group were higher than those of the chemotherapy（P〈0.01）.In conclusion,herbal Tanreqinginjection has certain protective effects on immune system of body with lung carcinoma treated by chemotherapy,and also possesses promotive effects on antitumor immunity.
