小鼠不同途径感染真菌模型的建立及LD50的测定

摘要 目的:建立一种稳定的真菌感染的小鼠模型。方法选用健康6~8周龄的BALB/c雌性小鼠,体重16~18g,选择静脉和鼻饲给药,每天进行死亡情况观察,连续观察26~31d,计算小鼠的半数致死量(LD50)。结果经尾静脉注射白假丝酵母菌给药小鼠高剂量组(2×10^7)全部死亡,中剂量组(2×10^6)4只死亡,低剂量组(2×10^5)没有死亡,鼻饲给药组小鼠高中低剂量均无死亡,说明小鼠的死亡与剂量高低、给药途径有直接关联的小鼠出现死亡。结论在利用小鼠做真菌感染的实验时,尽可能选择鼻饲给药。 Objective To obtain a stable mouse model of fungal infection .Methods Female BALB/c mice,6 to 8-weeks-old,weighing 16 to 18g,were used for 50% lethal dose ( LD50) evaluation of Candida albicans , which were injected intravenously and intranasally .Mice were kept under observation for 31 days and deaths were recor-ded on a daily basis .Results Mice with tail vein injection of Candida albicans in high dose group (2 ×10^7 ) all died, four death in the medium dose group(2 ×10^6),no death in low dose group(2 ×10^5).Nasal feeding mice had no death in all groups ,saying that dose and routes of administration were directly related to the death of mice .Conclusion The intranasal route provided a simple ,non-invasive murine model for investigating fungal infection .
