门静脉高压症apelin/APJ信号活化的实验研究

摘要 目的:探讨apelin/APJ信号在门静脉高压症的激活状态以及门静脉高压症的过程是否有apelin/APJ信号的活化,并提出其可能的机制。方法:采用部分门静脉结扎(partial portal vein ligation,PPVL)及胆道结扎离断(bile duct ligation,BDL)两种方法制备SD大鼠不同特点的门静脉高压症模型。通过大体观察、血流动力学、病理学及免疫组织化学评估门静脉压力和apelin/APJ信号在肠系膜的激活。结果:两种模型组大鼠门静脉压力均比相应假手术组高。两种模型肠系膜血管新生明显,其侧支循环大量开放。BDL组和PPVL组大鼠肠系膜组织APJ和apelin蛋白与各自假手术组相比均有升高。结论:apelin/APJ信号可能在门静脉高压症形成过程中有重要作用。 Objective To explore apelin/APJ signaling in the formation of portal hypertension and the activation of apelin/APJ signaling pathway in the process of portal hypertension. Methods Portal hypertension model was induced by two methods included partial portal vein ligation(PPVL) and bile duct ligation(BDL). Portal pressure and the activation of apelin/APJ signaling of mesentery were evaluated by general observation, hemodynamics, pathology and immunohisto- chemistry. Results The portal pressure of the two types of portal hypertension model group increased significantly com- pared with each sham-operated group. The formation of angiogenesis and collateral vessel of mesentery was increased in the rats of 2 models. The expression of APJ and apelin protein of the mesentery was increased in the rats of BDL and PPVL. Conclusions Apelin/APJ signaling may contribute to the formation of portal hypertension.
