摘要
目的探讨乙醛脱氢酶2(ALDH2)对大鼠百草枯中毒性急性肺损伤(ALI)的保护作用。方法 SD大鼠120只随机均分为五组。A组大鼠作为空白对照;B、C、D和E组以百草枯(80mg/kg)一次性灌胃建立百草枯中毒大鼠ALI模型,2h后按1×10-8 mol/L硫辛酸(C组)、大豆苷(D组)和硫辛酸+大豆苷(E组)灌胃;A、B组给予10%NaOH灌胃作对照。在中毒后6、24、48和72h每组分别取6只大鼠,检测血清超氧化物歧化酶(SOD)的活性和丙二醛(MDA)含量,并检测肺组织线粒体中ALDH2的活性。结果与A组相比,B、C、D和E组血清SOD活性均降低(P<0.05),降低幅度依次为E组>D组>B组>C组;B、C、D和E组血清MDA浓度均增高(P<0.05),且B、D、E组高于C组(P<0.05);B、C、D、E组肺组织线粒体中ALDH2活性均增高(P<0.05),增高幅度依次为C组>B组>D组>E组。结论 ALDH2对百草枯中毒性大鼠ALI有保护作用。
Objective To investigate the protective effect of aldehyde dehydrogenase 2 (ALDH2) on paraquat-induced acute lung injury(ALI) in rats. Methods A total of 120 SD rats was equally randomized into 5 groups. The rats of group A were taken as blank controls. ALI models were established by paraquat 80 mg/kg gavage once in groups of B, C, D and E, which were treated with gavage of 10 - NaOH, lipoie acid 1 X 10-8 mol/L, daidzin 1 × 10-8 mol/L and lipoic acid plus daidzin once a day at 2 hours after ALI model was induced. Six rats from each group were randomly chosen at 6, 24,48 and 72 hours for detecting the activity of superoxide dismutase (SOD) and malondialdehyde (MDA) content in serurrL The activity of ALDH2 in mitochondria of fresh lung tissue was examined as well. Results Compared with group A, the activity of serum SOD was decreased in groups of B,C, D and E(P〈0. 05). The decrease in activity of serum SOD was in an order of group E-group D- group B-group C. Compared with group A, serum MDA content was increased in groups of B, C, D and E(P〈0. 05), which was higher in groups of B, D and E than that in group C(P〈0. 05). Compared with group A, the activity of ALDH2 in mitochondria of fresh lung tissue was increased in groups of B,C,D and E(P〈0. 05). The increase in ALDH2 activity was in an order of group C〉 group B:〉group D-group E. Conclusion ALDH2 has a protective effect on paraquat-induced ALI in rats.
