摘要
目的:探讨海藻酸钙敷料对体外培养的成纤维细胞相对增殖率的影响,初步评价该材料的细胞毒性作用并探讨对细胞增殖影响最低的条件。方法:第一步:采用四唑盐(MTT)比色法检测不同浓度(0、50%和100%)敷料浸提液的吸光度,分别计算各浓度下成纤维细胞的相对增殖率,初步评价其细胞毒性。第二步:根据上述结果,继续采用MTT法检测细分浓度(0、10%、20%、30%、40%和50%)敷料浸提液对成纤维细胞增长率影响,测出对细胞增殖无影响的材料溶液浓度。结果:各浓度浸提液均出现较高相对细胞增殖率,其细胞毒性均为0或1级,无毒性。精检MTT结果提示,相对浸提液浓度低于10%海藻酸盐敷料浸提液对成纤维细胞的生长无任何影响,浓度高于20%的浸提液对成纤维细胞的生长开始产生影响,但仍未到毒性标准。结论:海藻酸钙敷料无细胞毒性,具有良好细胞相容性,满足人体植入材料体外细胞毒性实验要求,可作为安全的伤口敷料使用。
Objective: To detect the relative fibroblast proliferation rate effected by the alginate calcium dressing in vitro, and evaluate the cytotoxicity of the material and the worst environmentthat affected the proliferation of fibroblast. Methods: First step: Tetrazolium salt colorimetry(methyl thiazolyl tetrazolium, MTT) was used to detect the absorbance of the extracted liquid of the material in different concentrations (0, 50% and 100%), and then, the relative proliferation rate of fibrobtast in each group were calculated. Second step: The cytotoxicity was evaluated according to the result. The MTT was continued applicated to detect the absorbance of extracted liquid of the materials in more finely differentiated concentrations (0, 10%, 20%, 30%, 40% and 50%), to detect the lowest con- centration that begun to affect the proliferation of fibroblast. Results: The results of first step showed that the relative proliferation rate in each group kept up a high level, and the cytotoxicity of extracted liquid of material in each group was 0 or 1 grade,which has no cytotoxicity. The results of second step showed that the concentration under 10% had no influence at all on the proliferation of fibroblast. When the concentration was over 20%, proliferation of fiberblast was affected, but still in the range of non-cytoxicity. Conclusion: Alginate calcium dressing had no cytotoxicity, and pos- sessed a good cytocompatibility. It met the cytotoxicity demand to an implant material in vitro, and was desired to be an appropriate wound dressing.
