摘要
从富油土壤中分离筛选到一株脂肪酶产生菌M-4,通过对M-4进行形态和r DNA ITS序列鉴定,确定其属于酵母菌Sporidiobolus salmonicolor strain。通过正交试验设计对该菌株进行产酶条件探索,得出其摇瓶培养最适产酶条件为:种子培养时间为90 h,最佳发酵培养基碳源为葡萄糖2%+橄榄油3%,氮源为5%酵母膏,初始p H值为7.0,发酵温度为30℃,在此条件下的产酶可达12.2 U/m L。
The M-4 strain was achieved by screening from the soil near Guangzhou Institute of Energy Conversion (GIEC). This strain was identified as Sporidiobolus salmonicolor strain based on its morphology and rDNA ITS sequence analysis. The optimal lipase-producing conditions of M-4 were investigated conditions are optimized as: carbon source value is 7.0; the time of enzyme producing lipase productivity can reach 12.2 U/mL. is glucose 2%+ olive oil 3% ; is 90 h; optimal fermentation through orthogonal test.
