Expression of thymidylate synthetase gene in 64 primary hepatic cancer patients

摘要 目的分析胸苷酸合成酶(Thymidylatesynthetase,TYMS)基因在原发性肝癌中的表达及其临床意义。方法应用荧光定量PCR技术,对64例原发性肝癌TYMS基因进行扩增,免疫组化对照。结果在64例肝癌中有28例表达TYMS,占43.8%,拷贝数中位数M=3.55×10^4copies/mL,TYMS基因拷贝数对数(Logcopies)与免疫组化分值有相关性,r=0.97,其中高拷贝(〉1.0×10^4)表达有7例(10.9%),中拷贝((1.0~10)×10^4)表达有10例(15.6%),低拷贝(〈10×10^3)表达有11例(17.2%),TYMS的表达与肿瘤大小、生存期(术后1年)、是否癌旁组织差异有统计学意义(P〈0.05),而与病理类型、AFP水平、HBV—DNA差异无统计学意义(P〉0.05)。结论荧光定量PCR技术可替代免疫组化方法检测TYMS基因的表达,TYMS基因在原发性肝癌有表达,对肿瘤生长和转移中有促进作用,可作为原发性肝癌的靶向治疗证据。 Objective To analye the expression of thymidylate synthetase( TYMS ) gene in primary hepatic cancer cases. Methods Fluorescence quantitative PCR(FQ-PCR)was used to detect TYMS in 64 hepatocellular carcinoma cases and the results were ascertained with immunohistochemieal method . Results TYMSs was expressed in 64 primary liver cancer eases, occupied 43.8%, and the median copy number was M=3.55×10^4 copies/ml, being correlated with immunohistochemical score in TYMS gene copy number ( Log copies, r = 0.97) .High expression was noticed in seven cases (〉1.0× 10^5,accounted for 10.9%, moderate expression was noticed in 10 cases((1.0-10) × 10^4 ) accounted for 15.6% and low expression (〈10× 10^3) l 1 eases (17.2%) . The expression of thymidylate synthetase in primary hepatic cancer cases revealed significant differences in tumor size, survival ( 1 year after operation)and paracaneerous tissue (P 〈0.05) .But there were no significant differences were found in pathological type, AFP level and HBV-DNA in the cases(P 〉0.05) .Conclusion FQ-PCR technology would take the place of immunohistoehemieal method for detecting TYMS gene in primary liver cancer patient. TYMS gene plays a part in promoting growth and metastasis of hepatic carcinoma it and might be taken as evidence of targeted therapy of primary liver cancer.
