天麻配方颗粒高效液相色谱指纹图谱研究

HPLC characteristic chromatogram of Formula Granule of Gastrodiae Rhizoma

目的研究天麻配方颗粒的高效液相色谱指纹图谱,为其质量控制提供更可靠的方法。方法采用高效液相色谱法,色谱柱为Megres5-C18(250 mm×4.6 mm,5μm),以乙腈-0.05%磷酸溶液进行梯度洗脱,柱温为30℃,体积流量为0.8mL/min,检测波长为220 nm。结果初步建立了天麻配方颗粒的高效液相色谱指纹图谱,确定了6个特征峰。结论高效液相色谱指纹图谱可为天麻配方颗粒的鉴别和质量控制提供参考。

Objective To establish the HPLC characteristic chromatogram of Formula Granule of Gastrodiae Rhizoma as a reference for the identification and quality control of this formula granule. Methods The chromatograms were obtained using the Megres5-C18 column （250 mm×4.6 mm,5μm） with mobile phase comprised of acetonitrile-0.05% phosphoric acid aqueous solution in gradient elution at flow rate of 0.8 mL/min, column temperature 30 ℃, detection wavelength 220 nm and sample size of 10 μL. Results The HPLC characteristic chromatogram of Formula Granule of Gastrodiae Rhizoma consisted of 6 common peaks. The similarity of 10 batches of samples was above 0.9. Conclusion The HPLC method is proved to be accurate, stable and reproducible and can provide reference for the identification and quality control of Formula Granule of Gastrodiae Rhizoma.
