摘要
试验主要对ELISA定量测定苦荞制品中黄曲霉毒素B1的可行性进行研究,研究了ELISA法测定黄曲霉毒素B1的标准曲线、灵敏度、精密度、准确度以及对不同苦荞制品对检测结果的影响。结果表明,ELISA分析方法的检测范围为0.1μg/kg^5μg/kg,最低检测限量达到2μg/kg,批内和批间的变异系数均小于5%,在0.1μg/kg^5μg/kg范围的平均加标回收率为93.6%;苦荞制品基质对ELISA检测法的影响从小到大依次为苦荞粉、苦荞面和苦荞茶,当苦荞粉稀释倍数在2~30倍、苦荞面稀释到10倍、苦荞茶稀释到30倍能满足ELISA检测需求。试验证实采用ELISA检测苦荞制品中黄曲霉毒素B1具有可行性。
The experiment was performed to study the feasibility of determination of aflatoxin B1 in tartary buckwheat by ELISA, The result indicated that the detection range of ELISA analysis was 0.1 μg/kg-5 μg/kg. The detection sensitivity was to 0.1 μg/kg and the variations within, and between batches were all less than 5%. Furthermore, the average standard addition recoveries were 93.6% in the context of 0.1 μg/kg to 5 μg/kg. The ELISA analysis most affected by tartary buckwheat product matrix was buckwheat tea, followed by buckwheat powder and then buckwheat flour. To meet the needs of ELISA analysis, the dilution multiple of buckwheat powder, buckwheat flour and buckwheat tea were 2-30 times, 10 times and 30 times respectively. This experiment confirmed the determination of aflatoxin Bl in tartary buckwheat products by ELISA was feasible.
