摘要
建立了黑果枸杞中花色素苷的HPLC半定量方法。采用ODS-C18柱(250nm×4.6nm,5μm);流动相A:1%甲酸+0.1%三氟乙酸水溶液;B:15%甲醇乙腈溶液;梯度洗脱为0-20min,15%→30%B;20-40min,30%→60%B;40-50min,60%→80%B;50-70min,80%→100%B;70-80min,100%B。检测波长530nm,流速为1.0mL/min。标准品在0.0125-0.5mg/mL范围内线性良好,平均回收率为99.89%,RSD为2.08%,黑果枸杞鲜果花色苷含量为(399.81±12.02)mg/100g。
A high performance liquid chromatographic(HPLC) method was developed for semi-quantitative determination of anthocyanin from Lycium ruthenicum Murr.. The sample was separated by an Agilent ZORBAX Stable Bond SB-C18(25 0 mm×4.6 mm, 5 μm) column. The mobile phase consisted of 1% aqueous formic acid solution with 0.5% trifluoroacetic acid(A) and 15% methanol in Methyl Cyanides solution(B). Elution was performed with a linear gradient as follows: 0-20 min, B from 15% to 30%; 20-40 min, B from 30% to 60%; 40-50 min, B from 60% to 80%; 50-70 min, B from 80%→100% B; 70-80 min, 100% B. The UV detection wavelength was set at 530 nm and system flow rate was 1.0 mL /min. A good linearity was obtained in the range of 0.012 5-0.5 mg/mL. The average recovery was 99.89% and the RSD was 2.08%. The content of the anthocyanins from Lycium ruthenicum Murr. was(399.81±12.02) mg/100 g.The results indicated that the method was simple, sensitive and accurate, and could be applied in the determination of anthocyanin from Lycium ruthenicum Murr.
