HPLC法测定骨痈内消丸中绿原酸的含量

Content Determination of Chlorogenic Acid in Guyong Neixiao Pill by HPLC

目的：建立测定骨痈内消丸中绿原酸含量的方法。方法：采用高效液相色谱法。色谱柱为Eclipse XDB C18（250 mm×4.6mm,5μm）,流动相为乙腈-0.2%磷酸溶液（8∶92,V/V）,流速为1.0 ml/min,检测波长为327 nm,柱温为30℃。结果：绿原酸的进样量在0.035 1~0.702 4μg范围内与峰面积积分值呈良好线性关系（r=0.9999）;精密度、稳定性、重复性试验的RSD≤1.8%;平均加样回收率为98.37%,RSD=1.93%（n=9）。结论：本方法准确、快速,重复性好,可用于骨痈内消丸的质量控制。

OBJECTIVE： To establish a method for the content determination of chlorogenic acid in Guyong neixiao pill. METHODS： HPLC method was adopted. The separation was performed on a Eclipse XDB C18（250 mm×4.6 mm, 5μm） column with mobile phase consisted of acetonitrile-0.2 % phosphonic acid （8：92, WV）at the flow rate of 1,0 ml/min. The detection wave- length was set at 327 nm, and column temperature was 30℃. RESULTS： The linear range of chlorogenic acid was 0.035 1-0.702 4 μg（r=0.999 9）o RSDs of precision, stability and reproducibility tests were all lower than 1.8%. The average recovery of 98.37% （RSD= 1.93%, n=9）. CONCLUSIONS： The method is accurate and rapid with good repeatability. It could be used for the quality control of this preparation.
