摘要
本试验以体外培养的单层Caco-2细胞为研究对象,以细菌易位数量(病原菌感染细胞的数量)和肠绒毛损伤程度(胞外乳酸脱氢酶LDH释放量)作为指标,研究蒙脱石-枯草芽孢杆菌(MMT-B.subtilis)复合物对肠黏膜屏障功能的影响效果。结果表明:最适宜Caco-2细胞生长的MMT-B.subtilis浓度是0.2 mg/m L;同MMT与B.subtilis的物理混合物相比,MMT-B.subtilis大大减少了病原菌入侵Caco-2细胞的菌落数,且差异显著(P<0.05);大肠杆菌和鸡白痢沙门氏菌严重破坏了Caco-2细胞膜的完整性,导致胞内LDH大量逸出释放到胞外,同时也显著抑制了Caco-2胞内的LDH活性,而加入了MMT-B.subtilis后,对Caco-2细胞内的LDH活性没有显著影响,同时没有破坏Caco-2细胞膜的完整性。结果表明,MMT-B.subtilis对细胞膜的保护作用强于MMT与B.subtilis混合物。
In this experiment, the Caco-2 cell monolayer was used as model, with the amount of bacterial translocation(cell number of pathogen infection) and intestinal villus damage(extracellular lactate dehydrogenase release quantity)as indicator, to study the effect of MMT-B.subtilis on intestinal mucosa. The results showed that the most appropriate concentration of MMT-B.subtilis for Caco-2 cell growth was 0.02 mg/L; Compared with the physical mixture, MMT-B.subtilis greatly reduced the number of colonies of pathogen invasion on Caco-2 cell and the difference(compared with the control group) was significant(P0.05); E. coli and S. pullorum seriously undermined the integrity of Caco-2 cell membrane, resulted that the LDH escaped from intracellular and released into the extracellular, and also inhibited the LDH activity of Caco-2 intracellular. After adding the MMT- B.subtilis, the Caco-2 LDH activity of intracellular was not affected, and overall, the protective effect of MMT- B.subtilis on cell membrane is stronger than the physical mixture of MMT and B.subtilis.
