摘要
目的本文旨在探讨转化生长因子β-1(transform growth factorβ1,TGF-β1)在高白细胞急性髓系白血病(acute myeloid leukemia,AML)发病中的作用。方法分别采用实时定量PCR、流式细胞术及酶联免疫反应检测38例AML患者骨髓单个核细胞(bone marrow mononuclear cells,BMMNC)内TGF-β1mRNA浓度、48例AML患者BMMNC的TGF-β1蛋白和40例AML患者血清TGF-β1蛋白质量浓度。结果高白细胞AML患者、非高白细胞AML患者和正常对照者的TGF-β1mRNA浓度差异无统计学意义,高白细胞和非高白细胞AML患者的胞内TGF-β1蛋白质量浓度显著高于正常对照者,血清TGF-β1蛋白质量浓度显著低于正常对照者。高白细胞和非高白细胞AML患者之间的胞内和血清TGF-β1蛋白质量浓度差异无统计学意义。高白细胞与非高白细胞AML患者之间胞内及血清TGF-β1质量浓度与FAB亚型无关。结论 AML患者血清TGF-β1浓度降低可能参与了疾病的发生,而血清TGF-β1的减少可能并不能反映BMMNC合成TGF-β1减少。TGF-β1对高白细胞AML的发病并未产生重要影响。
Objective To investigate the role of transform growth factor β1( TGF-β1) in pathogenesis of hyperleukocytic acute myeloid leukemia( AML). Methods The mRNA transcription levels of TGF-β1from bone marrow mononuclear cells( BMMNC) in 38 AML patients,protein expression levels of intracellular TGF-β1from BMMNC in 48 AML patients,and serum levels of TGF-β1from 40 AML patients were measured by real-time fluorescence quantitative polymerase chain reaction( RQ-PCR),flow cytometry,and doubleantibody sandwich enzyme-linked immunosorbent assay( ELISA),respectively. Results The relative levels of the TGF-β1gene expression among hyperleukocytic AML patients,non-hyperleukocytic AML patients,and NC has no significant difference( P 0. 05).The protein expression levels of TGF-β1in both hyperleukocytic AML and non-hyperleukocytic AML patients were significantly higher than that in NC( P 0. 05),the plasma levels were significantly lower than that in NC( P 0. 05). There was no significant difference in intracellular protein expression levels and plasma levels of TGF-β1between hyperleukocytic AML and non-hyperleukocytic AML patients( P 0.05). There was no significant difference in intracellular protein expression levels and plasma levels of TGF-β1between the FAB subtypes in hyperleukocytic and non-hyperleukocytic AML( P 0. 05). Conclusion Decreased serum levels of TGF-β1may be involved in the pathogenesis of AML. However,decreased TGF-β1serum levels may not reflect reduced TGF-β1production by BMMNC. Our study showed that TGF-β1had no important impact on the pathogenesis of hyperleukocytic AML.
