摘要
目的分析并鉴定大鼠肌肉注射胆木注射液后的主要入血的成分,初步探究其药效物质。方法采用UPLCPDA-QTOF/MS分析空白血浆、含胆木注射液血浆、胆木注射液和对照品,对比图谱中色谱峰保留时间以及离子碎片信息,解析成分的归属。结果以胆木注射液11个化学成分为参照,从大鼠血浆中鉴定了8个原形入血成分,分别是naucleamide A-10-O-β-D-glucopyranoside、短小蛇根草苷、3-表短小蛇根草苷、3α,5α-tetrahydrodeoxycordifoline lactam、naucleoxoside A、naucleoxoside B、异长春花苷内酰胺、喜果苷。结论 8个入血成分均为生物碱类成分,提示生物碱是胆木注射液可能的活性物质。
AIM To analyze and identify the constituents in rat plasma after intramuscular administration of Danmu Injection (Nauclea officinalis Pierre ex Pitard),and explore their active materials.METHODS UPLCPDA-QTOF/MS analysis was performed on Danmu Injection,rat plasma with and without Danmu Injection and references.The constituents in rat plasma were identified by comparing the retention time and mass spectrometry fragment data.RESULTS On the basis of eleven constituents of Danmu Injection,eight constituents found in their original forms in rat plasma were identified as naucleamide A-10-O-β-D-glucopyranoside,pumiloside,3-epi-pumiloside,3α,5α-tetrahydrodeoxycordifoline lactam,naucleoxoside A,naucleoxoside B,strictosamide and vincosamide.CONCLUSION The finding of alkaloids in rat plasma should suggest that alkaloids in Danmu Injection may be the active materials.
