腊梅花中黄酮类化合物的UHPLC/QTOF-MS分析 被引量:26

摘要 目的采用液相色谱-四极杆-飞行时间质谱技术UHPLC/QTOF-MS对腊梅花乙醇提取物中的黄酮类化合物进行分析鉴定。方法色谱分离采用Agilent Eclipse XDB-C18色谱柱(100 mm×2.1 mm,1.7μm),0.1%甲酸水溶液-乙腈梯度洗脱。质谱分析采用电喷雾离子源,正离子模式检测。结果通过二级质谱裂解分析以及参考文献,从腊梅花中鉴定出了绿原酸、金丝桃苷、异槲皮苷、槲皮素-3'-葡萄糖苷、山柰酚-3-O-半乳糖苷、紫云英苷、槲皮苷、槲皮素、山柰酚和异鼠李素等1个多酚类和9个黄酮类化合物。结论本法为腊梅花黄酮类化合物的分析鉴定提供快速和准确方法。 AIM To use ultra performance liquid chromatography/time-of-flight mass spectrometry (UHPLC/TOF-MS) to identify the flavonoids in ethanolic extract of Chimonanthus praecox (L.) Link.METHODS Chromatographic separation was performed on the Agilent Eclipse XDB-C18 column (100 mm × 2.1 mm,1.7 μm) and eluted with the mobile phase consisted of water containing 0.1% formic acid and acetonitrile in a gradient manner.Electro spray ionization source was used for the mass spectrometric analysis under positive ion mode.These components were further analyzed by MS^2 spectra.RESULTS By second mass spectrometric analysis in combination with the literature data,constituents in Chimonanthus praecox (L.) Link were identified as one polyphenol and nine flavonoids,including chlorogenic acid,hyperoside,quercetin-3-O-glucoside,quercitrin-3'-glucoside,kaempherol-3-O-galactoside,kaempherol-3-O-glucoside,quercetin-3-O-rhamnoside,quercetin,kaempferol,and isorhamnetin.CONCLUSION The method provides good and rapid qualitative means for the identification of flavonoids from Chimonanthus praecox.
