摘要
目的 HPLC法测定湖南省桂东县杜仲GAP基地不同品种和不同采收期杜仲样品中松脂醇二葡萄糖苷的量.方法 杜仲三氯甲烷提取液分析采用ZORBAX Eclipse XDB-C18色谱柱(4.6 mm×250 mm,5μm),流动相为甲醇-水(25∶75),检测波长为277 nm,体积流量为1.0 mL/min,柱温为25℃,进样量为5μL.结果 松脂醇二葡萄苷在0.49 ~4.90 μg范围内线性关系良好,r^2=0.999 9,其平均回收率为100.4%,RSD=2.1%.10年生以上的光皮杜仲和粗皮杜仲所含松脂醇二葡萄糖苷均高于0.10%,但前者高于后者.结论 推荐光皮杜仲为选优和繁殖的品种.
AIM To use HPLC method to measure pinoresinol diglucoside content from different cultivars and picking time of Eucommia ulmoides collected from GAP plantation in Guidon County,Hunan Province.METHODS The HPLC analysis of chloroform extract of E.Ulmoidescolumn was performed on ZORBAX Eclipse XDBC18 (4.6 mm ×250 mm,5 μm) with the mobile phase consisted of methanol-water.The detective wavelength was set at 277 nm,the column temperature was 25 ℃,the flow rate was 1.0 mL/min,and the injection size was 5 μL.RESULTS The calibration curve of pinoresinol diglucoside was linear in the range of 0.49-4.90 μg (r^2 =0.999 9),its average recovery was 100.4% (RSD =2.1%).The pinoresinol diglucoside content of the smooth skin and the rough skin E.ulmoides were all over 0.10%,but the former was higher than the latter.CONCLUSION The smooth skin E.ulmoides should be recommended as the cultivar.
