摘要
目的从细胞水平研究灵芝多糖对B16F10黑素瘤细胞分泌TGF-β1的影响,探讨灵芝多糖拮抗B16F10黑素瘤细胞分泌TGF-β1分子从而治疗黑素瘤的可能机制。方法用不同浓度灵芝多糖处理B16F10黑素瘤细胞,并设立无灵芝多糖的对照组。采用Western blot、RT-PCR和免疫组化的方法检测灵芝多糖作用48h后B16F10黑素瘤细胞表达TGF-β1的水平。结果 Western blot法、RT-PCR法和免疫组化法均显示B16F10黑素瘤细胞TGF-β1的表达量明显低于对照组(P<0.05),且随着灵芝多糖浓度的增加而降低更加明显。结论灵芝多糖可拮抗B16F10黑素瘤细胞分泌TGF-β1,起到抑制B16F10黑素瘤进展的作用。
Objective To determine the effect of Ganoderma lueidum polysaccharides(G1-PS) on the expression of TGF-[31 in B16FlO melanoma cells in vitro, and to explore its possible mechanism that G1-PS treats melanoma. Methods B16FlO melanoma cells were treated with or without different concentrations of G1-PS for 48h. The expression levels of TGF-β1 were assessed by Western-blot,RT-PCR and Immunocytochemistry. Results Western-blot, RT-PCR and immunocytochemistry showed that compared with normal controls, the expression levels of TGF-β1 significantly decreased in B16F10 melanoma cells treated with Gl-PS for 48h (P 〈 0.05 ),and the relative expression levels of TGF-β1 decreased progressively as the concentration of Gl-PS increased. Conclusion Gl-PS can effectively decrease the expression of TGF-β1 in B16F10 melanoma cells and possibly inhibits the progression of B16F10 melanoma.
