摘要
[目的]得到清晰的特异性条带,更好地分析SRAP标记产物。[方法]对聚丙烯酰胺电泳的上样量、电压条件进行了梯度试验,同时就灌胶方式、银染时间、显影剂调配等各步骤的操作进行了详细地探讨。[结果]上样量7μl、电压1 000 V最合适,能得到DNA条带清晰、特异性条带区分效果明显的条带。[结论]为后续电泳操作及分析过程奠定技术基础。
[Objective] The aim was to get clear specific bands,in order to better analyze SRAP markers.[Method] Sample amount and voltage condition of polyacrylamide electrophoresis were tested.at the same time,glue,silver dyeing time,the developer deployment and so on were made a detail discussion on the operation of the flash.[Result] 7 μl of sample amount,1 000 V of voltage was the most appropriate,and coult get clear DNA banding and specificity belt to distinguish the effect of belts.[Conclusion] The study laid a solid technical foundation for convenient operation and subsequent analyses process.
