独流减河野生鲫鱼的同工酶分析 被引量:2

摘要 为了解天津市独流减河野生鲫鱼的遗传多样性,采用水平淀粉凝胶电泳法检测采自47尾二倍体野生鲫鱼的7种同工酶和肌浆蛋白,包括AAT、GPI、IDH、LDH、MDH、ME、PGM和PROT.共检测出18个基因座位,有变异的基因座位为AAT-3*、GPI-1*、GPI-2*、GPI-3*、IDH-2*、ME*、PGM*和PROT*;同工酶GPI的3个基因座位GPI-1*、GPI-2*、GPI-3*均含有3个等位基因,AAT-3*、IDH-2*、PGM*、PROT*和ME*含有2个等位基因;平均杂合度观察值为0.071 5,平均杂合度预期值为0.076 7;χ2检验表明有变异的基因座位均符合哈代-温伯格平衡.这些结果表明天津市独流减河野生鲫鱼具有丰富的遗传多样性. To investigate the genetic diversity of wild crucian carp collected from Duliujian river of Tianjin, 7 kinds of isozyme and myogen from forty-seven wild crucian carp, namely AAT, GPI, IDH, LDH, MDH, ME, PGM and PROT, were analyzed by horizontal starch-gel electrophoresis. Eighteen gene loci were detected, and the loci of AAT-3*, GPI-1*, GPI-2*, GPI-3*, IDH-T, ME*, PGM* and PROT* were polymorphic. The loci of GPI-1*, GPI-2* and GPI-3* all had three alleles, and the loci of AAT-3*, IDH-2*, ME*, PGM* and PROT* all had two alleles. The mean heterozygosity observed was 0.071 5 and the value expected was 0.076 7. Thex2 test indicated that polymorphic loci conformed with Hardy-Weinberg equilibrium. The results showed that wild crucian carp from Duliujian river had rich genetic diversity.
