二甲双胍对糖尿病非酒精性脂肪性肝病大鼠G-6-Pase表达的影响

摘要 目的观察二甲双胍对糖尿病非酒精性脂肪性肝病大鼠G-6-Pase表达的影响。方法 SD大鼠(雄性)随机分为正常对照组(10只),肥胖组(12只),糖尿病组(10只),二甲双胍治疗组(9只)。采用RT-PCR方法测定4组大鼠肝组织中葡萄糖6磷酸酶(G-6-Pase)mRNA表达,并测定各组空腹血糖(FBG)、甘油三酯(TG)、总胆固醇(TC)、游离脂肪酸(FFA)及空腹胰岛素(Fins)。结果糖尿病组及糖尿病二甲双胍控制组大鼠肝脏G-6-Pase mRNA均较肥胖组明显增高(P<0.05),糖尿病二甲双胍控制组G-6-Pase mRNA较糖尿病组明显降低(P<0.05)。肝HE染色糖尿病二甲双胍控制组大鼠肝脏脂肪变性程度减轻。结论二甲双胍可改善大鼠糖尿病非酒精性脂肪性肝病的肝脏病理学损害,其机制可能与G-6-Pase下调有一定关系。 Objective To observe the effect of metformin on the expression of G-6-Pase in diabetic rats with nonalcoholic fatty liver disease.Methods Male Sprague-Dawley rats were randomly divided into four groups:normal control group( n = 10),obesity group( n = 12),diabetes mellitus group( n = 10),and diabetes controlled by metformin group( n = 9).The G-6-pase mRNA expression in liver tissue was measured by RT-PCR.The blood samples were measured for blood glucose,lipid,and FFA biochemically,and insulin by radio-immunoassay.Results Expressions of G-6-Pase mRNA in diabetes mellitus group,diabetes controlled by metformin group were significantly higher than obesity group( P〈0.05);G-6-Pase mRNA of diabetes treated with metformin was significantly lower than diabetes mellitus group( P〈0.05),The expression of G-6-Pase mRNA of obesity group was significantly lower than normal control group( P〈0.01);Liver fatty degeneration was mild as compared with model group.Conclusion Metformin could improve liver pathology damage in diabetic rats with nonalcoholic fatty liver disease,which appears to be associated with decreased G-6-Pase mRNA.
