摘要
目的探讨瞬时感受器电位香草酸受体3离子通道蛋白(TRPV3)在膀胱癌细胞中的mRNA表达水平,以及TRPV3激动剂Thymol对膀胱癌细胞增殖和迁移能力的影响。方法采用实时荧光定量PCR检测TRPV3 mRNA在膀胱癌细胞系RT4,UM-UC-3,SW780,J82,TCC-SUP,T24和人膀胱上皮永生化细胞系SV-HUC-1中的表达;利用Thymol激活膀胱癌细胞TRPV3通道,并用WST-1和细胞划痕实验分别检测Thymol激活TRPV3后膀胱癌细胞的增殖和迁移能力变化。结果相较于正常人膀胱上皮细胞SV-HUC-1,各种膀胱癌细胞系的TRPV3 mRNA表达水平均明显下调(P<0.0001)。与不加激动剂的DMSO对照组比较,加入Thymol激活膀胱癌细胞TRPV3通道后,膀胱癌细胞的增殖和迁移能力明显降低(P<0.01)。结论 TRPV3基因在膀胱癌细胞中表达明显降低,激活TRPV3通道后可明显减慢膀胱癌细胞增殖和迁移速度。
Objective The Transient receptor potential cation channel,subfamily V,member 3( TRPV3) is a nonselective cation channel protein.Aim of our study was to investigate TRPV3 mRNA expression and function in human bladder carcinoma cells.Methods Real-time RT-PCR was performed to detect TRPV3 mRNA expression in bladder carcinoma cells compared with normal human urothelium cells.Furthermore,the function of TRPV3 in bladder carcinoma cells was detected by WST-1 and cell wound scratch assay.Results TRPV3 gene was low-expressed in bladder carcinoma cells and TRPV3 agonist Thymol obviously inhibited the proliferation and migration of bladder carcinoma cells.Conclusion TRPV3 was down-regulated in bladder carcinoma cells and its agonist suppressed the proliferation and migration of bladder carcinoma cells,suggesting that TRPV3 plays a role in the development of bladder carcinoma.
