葡萄球菌属16种黏附素编码基因研究

摘要 目的调查一组凝固酶阳性葡萄球菌黏附素编码基因携带状况,分析黏附素编码基因及荚膜抗原编码基因检出率。方法收集2013年3-9月江苏省江阴市人民医院住院患者标本中分离的凝固酶阳性葡萄球菌共20株,用spa基因PCR检测作金黄色葡萄球菌的分子鉴定,再采用聚合酶链反应(PCR)的方法分析16种黏附素编码基因和荚膜抗原的型别编码基因(cap5、cap8)。结果 20株凝固酶阳性葡萄球菌中13株为金黄色葡萄球菌,其余7株为非金黄色葡萄球菌;13株金黄色葡萄球菌黏附素编码基因每一株均有阳性检出,除sasX基因阳性率低外,其余均较高;荚膜抗原编码基因12株有阳性检出;7株非金黄色葡萄球菌无任何黏附素与荚膜抗原编码基因检出。结论对金黄色葡萄球菌进行16种黏附素编码基因及2种荚膜抗原编码基因联合检测研究尚为国内外首次,13株金黄色葡萄球菌黏附素编码基因及荚膜抗原编码基因高检出率可能与菌株定植及感染相关。 OBJECTIVE To investigate the distribution of the genes encoding adhesin in a group of Staphylococcus spp and analyze the detection rate of the genes encoding adhesin and the genes encoding capsular antigen . METHODS A total of 20 strains of coagulase‐negative Staphylococcus were isolated from the specimens that were obtained from the hospitalized patients who were treated in the Jiangyin People's Hospital from Mar 2013 to Sep 2013 ,then the molecular identification of Staphylococcus aureus was performed by using spa PCR ,and the 16 genes encoding adhesion and the genes encoding capsular antigen (cap5、cap8) were analyzed by means of polymer‐ase‐chain‐reaction (PCR) .RESULTS Of the 20 strains of coagulase‐negative Staphylococcus ,13 strains were the S .aureus ,and the rest of 7 strains were non‐S .aureus;each of the 13 strains of S .aureus was tested positive for the gene encoding adhesin ,and the positive rate of all the encoding genes were high except for sasX gene;the genes encoding capsular antigen were tested positive in 12 strains;neither the genes encoding adhesin nor encoding capsular antigen were detected in the 7 strains of non‐S .aureus .CONCLUSION It is the first report that the 16 genes encoding adhesin and 2 genes encoding capsular antigen are detected in the S .aureus strains .The positive rates of the genes encoding adhesin or capsular antigen are high in the 13 strains of S .aureus ,which may be asso‐ciated with the bacterial colonization or the infections .
