黄花绿绒蒿与蓝花绿绒蒿的比较研究 被引量:1

摘要 目的通过黄花绿绒蒿与蓝花绿绒蒿的药材性状、解剖特征、化学组分的TLC定性与HPLC定量的比较,拟建立黄花绿绒蒿的药材质量标准和蓝花绿绒蒿的鉴定技术指标。方法采集对口全缘绿绒蒿、总状绿绒蒿、五脉绿绒蒿、川西绿绒蒿进行性状、解剖特征比较。用TLC定性分析,确定其药用单体指标成分,并用HPLC定量分析其指标成分。结果四种绿绒蒿均含木犀草素,HPLC的色谱条件为色谱柱:Chrompsuper C18(4.6mm×250mm,5μm);流动相:甲醇-0.2%磷酸溶液(60:40),流速0.6L·min-1;柱温30℃;检测波长349nm;木犀草素在0.084μg^0.84μg范围内呈良好的线性关系(R2=0.9995),平均加样回收率为97.48%,RSD为0.91%(n=5)。结论全缘绿绒蒿为藏药黄花绿绒蒿,木犀草素的含量为0.013%,可作为其药材质量的控制指标之一。总状绿绒蒿、五脉绿绒蒿、川西绿绒蒿为蓝花类绿绒蒿,其TLC、HPLC图谱均与全缘绿绒蒿有明显差异,因此,黄花绿绒蒿与蓝花类绿绒蒿药材不应混用。 Objective To establish the quality standard for crude drug of yellow flowers Meconopsis and the technical specification of identification of blue flowers Meconopsis by comparing their drug properties,anatomical characteristics,TLC identification and HPLC quantitative determination.Methods The drug properties and anatomical characteristics of M.integrifolia,characteristics of M.integrifolia,M.racemosaM.henrici and 31.quintuplinervia were compared,themedicinal monomer constituent was confirmed by TLC and quantitatively determined by HPLC.Results Four kinds of Meconopsis all have luteolin.The condition of HPLC:Chrompsuper C1s column (4.6mm×250μm, 5gm) and methanol -0.2% orthophosphoric acid(60:40 ) as the mobile phase,the flow rate was 0.6L·min-1,and the column temperature was 30 ℃,the detection wavelength was 349nm ,the linear range of luteolin was 0.084μg ~ 0.84 ( R2=0.999 5), the average recovery was 97.48% with the RSD was 0.91%(n=5).Conelusion As yellow flowers Meconopsis,the contents of luteolin in M.integrifolia was 0.013%,It can be used to evaluate the quality of M.integrifolia.Mracemosa,Mhenrici and Mfuintup/inerria, as Blue Flowers Meconopsis,their TLC and HPLC have obvious differences from M. integrifgolia.So yellow flowers Meconopsis and blue flowers Meconopsis can not be mixed as drug.
