生物信息模拟及生物学效应结合研究盐酸小檗碱对乙酰胆碱酯酶的抑制作用 被引量:1

摘要 目的体外研究盐酸小檗碱对乙酰胆碱酯酶(AchE)活性的抑制作用。方法利用生物信息模拟技术,体外模拟以盐酸小檗碱为代表的5种生物碱(配体)与AchE(受体)相互分子对接效应与差异性;将小鼠眼眶取血后脱臼处死,立刻开腹取出十二指肠制备组织匀浆,采用双缩脲法测定肠组织匀浆中总蛋白含量,将盐酸小檗碱与肠组织液共孵育后用酶法检测小鼠十二指肠组织和血清中AchE活性,以生物信息模拟和生物学效应相结合对比研究盐酸小檗碱对AchE的抑制作用。结果体外分子模拟5种生物碱中只有盐酸小檗碱和药根碱能与AchE受体活性中心进行分子对接;盐酸小檗碱能明显抑制小鼠十二指肠组织和血清中AchE活性,抑制程度与其浓度具有明显的量效关系。结论体外生物信息模拟和生物学效应研究均表明盐酸小檗碱对AchE有明显的抑制作用。 Objective To examine the inhibiting effect of berberine hydrochloride on acetylcholinesterase (AchE)in vitro.Methods The biological information simulation technology was used to simulate the molecular docking effects between 5 alkaloids(ligand)and AchE(receptor)in vitro and the difference of the effects was compared.The mice were sacrificed after the blood was obtained from the eye orbit area and the duodenum was removed to prepare tissue homogenate.The protein contents of the tissue homogenate were determined by using biuret test.The activity of AchE in both the duodenum and serum was detected by using enzyme test after incubation with berberine hydrochloride.The biological information simulation and biological effects were used to analyze the inhibiting effects of berberine hydrochloride on AchE in vitro.Results Among the five alkaloids,only berberine hydrochloride and jatrorrhizine could make molecular docking with the active center of AchE.Berberine could significantly inhibit the activity of AchE in both the duodenum and serum in a concentration-dependent manner.Conclusion Both of the in vitro biological information simulation technology and biological effect study showed that berberine hydrochloride had a significant inhibiting effect on AchE.
