白芍总苷脂质体对荷瘤小鼠肿瘤生长及免疫功能的影响 被引量:31

Effects of liposomes of total glucosides of paeonia on tumor growth and immune function in tumor-bearing mice

摘要 目的:探讨白芍总苷脂质体的体内抗肿瘤作用及其对荷瘤小鼠免疫功能的影响。方法:建立小鼠肉瘤S180及小鼠肝癌H22移植性肿瘤模型,观察白芍总苷脂质体对荷瘤小鼠肿瘤生长的抑制作用;采用腹腔巨噬细胞吞噬试验、淋巴细胞转化试验及western blot检测细胞因子IL-2,IL-12,TNF-α的蛋白表达,探讨白芍总苷脂质体对荷瘤小鼠免疫功能的影响。结果:在50,100,200 mg·kg-1时白芍总苷脂质体对S180的抑瘤率分别为33.51%,47.03%,57.83%,对H22的抑瘤率分别为25.53%,41.96%,49.22%,且能提高荷瘤小鼠的脾指数和胸腺指数;白芍总苷脂质体能增强荷瘤小鼠腹腔巨噬细胞的吞噬功能,促进淋巴细胞转化反应,上调荷瘤小鼠细胞因子IL-2,IL-12,TNF-α的蛋白表达。结论:白芍总苷脂质体具有较强的体内抗肿瘤活性,其机制可能与增强荷瘤小鼠的免疫功能有关。 Objective:To study the effect of liposomes of total glucosides of paepnia (TGP liposomes) on the tumor growth and immune activity in tumor-bearing mice. Methods : The antitumor activity of TGP liposomes in vivo was observed in implanted H22 and S180 tumor models in mice. The effect of TGP liposomes on immune function in tumor-bearing mice was investigated by measuring the expression levels of IL-12, IL-2 and TNF-α with phagocytosis test, T lymphocyte proliferation test and Western blot. Results: At dosages of 50, 100, and 200 mg kg-1, the inhibition rates of TGP liposomes were 33.51% , 47.03% and 57.83% for Slso, 25.53% , 41.96% and 49.22% for H22, respectively. TGP liposomes could increase spleen index and chest gland index, improve the phagocytosis function of the macrophage, enhance the T lymphocytes proliferation and increase the protein expres- sion levels of IL-2, IL-12 and TNF-α. Conclusion: TGP liposomes exhibited significant anti-tumor effect in vivo and the mechanism may be related to enhancing immune function.
