中国弱精症患者精子DJ-1蛋白与活性氧的关系 被引量:2

Relationship between reactive oxygen species and DJ-1 in ejaculated spermatozoa from Chinese asthenozoospermia patients

摘要 目的:探讨弱精症患者精子DJ-1蛋白与活性氧的关系。方法:分别收集349例弱精症患者和197例年龄相仿的健康志愿者正常精液样本,采用常规方法分离精子,免疫印记方法检测精子细胞DJ-1蛋白表达,并应用荧光显微镜和流式细胞仪检测其活性氧(reactive oxygen species,ROS)含量。结果:弱精症患者组的精子细胞中DJ-1蛋白表达明显下调,同时ROS含量增加。结论:弱精症患者精子DJ-1蛋白低表达伴随着活性氧含量的增加,导致氧化应激损伤、精子运动能力下降,是弱精症可能的发病分子机制之一。 Objective: To explore the relationship between reactive oxygen species and DJ-1 in ejaculated spermatozoa from Chinese asthenozoospermia patients. Methods: Totally 349 semen samples of asthenozoospermia patients and 197 semen samples of age-matched healthy volunteers were collected. Then sperm cells were separated with a conventional method, the DJ-1 level was measured with Western blotting, and reactive oxygen species (ROS) was evaluated by fluorescence microscopy and flow cytometry. Results: For the samples of asthenozoospermia patients, D J-1 decreased considerably, and the reactive oxygen species significantly increased compared with the normal group. Conclusion: There is a high possibility that the uncontrolled reactive oxygen species caused by DJ-l-downexpression in asthenozoospermia patients is one of the molecular mechanisms of the decreased sperm motility and asthenozoospermia.
