摘要
目的:建立HPLC同时测定脱脂紫苏子中迷迭香酸和木犀草素含量的方法。方法:采用Diamonsil C18色谱柱(4.6 mm×250 mm,5μm),流动相甲醇-0.1%磷酸溶液(55∶45),流速1 m L·min^-1,检测波长330,350 nm,柱温30℃。结果:迷迭香酸在9.91-158.55 mg·L^-1回归方程为Y=108.83X+0.14(r=0.999 9),平均回收率为99.68%,RSD 2.01%;木犀草素在0.997-15.952 mg·L^-1回归方程为Y=119.63X-3.68(r=0.999 9),平均回收率为100.34%,RSD 2.86%。结果:该方法操作简便、准确,重复性良好,可用于测定脱脂紫苏子中迷迭香酸和木犀草素的含量。
Objective: The aim of this study was to establish an HPLC method for simultaneous determination of the contents of Rosemary acid and Luteolin in defatted Perilla. Method: Diamonsil C18( 4. 6mm × 250 mm,5 μm) column was used,with methanol-0. 1% phosphoric acid( 55 ∶ 45) as mobile phase. The flow rate was set at 1. 0 m L·min^-1,the detection wavelength was set at 330 and 350 nm,with the column temperature at 30 ℃. Result: The linear response ranges were 9. 91-158. 55 mg·L^-1with regression curve Y =108. 83 X + 0. 136 8( r = 0. 999 9) for rosemary acid,and 0. 997-15. 952 mg · L^-1with regression curve Y =119. 63X- 3. 677 8( r = 0. 999 9) for luteolin. The average recovery of rosemary acid and luteolin were 99. 68%and 100. 34%,respectively( RSD 2. 01%,2. 86%). Conclusion: A simple,accurate and reproducible HPLC method has been provided for simultaneous determination of rosemary acid and luteolin in defatted Perilla.
