摘要
目的:观察秦皮醇提物(EEFC)对D-半乳糖所致亚急性衰老小鼠的抗衰老作用。方法:将ICR小鼠60只随机分为空白组(生理盐水100 mg·kg-1),模型组(D-半乳糖100 mg·kg-1),阳性药维生素E组(D-半乳糖100 mg·kg-1+维生素E30 mg·kg-1),EEFC低剂量组(D-半乳糖100 mg·kg-1+秦皮醇提物37.5 mg·kg-1)、中剂量组(D-半乳糖100 mg·kg-1+秦皮醇提物75 mg·kg-1)和高剂量组(D-半乳糖100 mg·kg-1+秦皮醇提物150 mg·kg-1),每组10只。连续ig给药60 d,末次给药后摘眼球取血,之后取脑、胸腺、脾脏、肝、皮肤,测定脑、胸腺、脾脏的脏器指数;测定血清、脑、肝、皮肤超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、过氧化氢酶(CAT)、谷胱甘肽过氧化氢酶(GSH-Px)、丙二醛(MDA)、总抗氧化能力(T-AOC)。结果:模型组的脑、胸腺、脾脏指数比空白组明显降低(P<0.05,P<0.01),小鼠血清、肝脏、皮肤、脑模型组T-AOC,SOD,CAT的值与空白组明显降低,MDA升高。秦皮醇提物各剂量组与模型组相比,高剂量组的脑、胸腺、脾脏质量指数显著增加(P<0.05,P<0.01)。秦皮醇提物组血清、脑组织、肝和皮肤中的SOD,CAT活性均显著增强(P<0.05,P<0.01),MDA含量降低(P<0.05,P<0.01),T-AOC显著增强(P<0.05,P<0.01),血清和皮肤中的GSH-Px活性显著增强(P<0.05,P<0.01)。结论:秦皮醇提物具有延缓D-半乳糖所致亚急性衰老小鼠的衰老作用。
Objective: To investigate the anti-aging effect of ethanol extract of Fraxini Cortex( EEFC) in subacute aging mice. Method: Sixty ICR mice were randomly divided into the blank group( physiological saline100 mg·kg^-1),the model group( D-galactose 100 mg·kg^-1),the positive group( D-galactose 100 mg·kg^-1+vitamin E 30 mg·kg^-1),the low dose group D-galactose 100 mg·kg^-1+ EEFC 37. 5 mg·kg^-1),the dose group( D-galactose 100 mg·kg^-1+ EEFC 75 mg·kg^-1),the high dose group( D-galactose 100 mg·kg^-1+ EEFC 150mg·kg^-1),each group of 10. And mice were treated by intragastric administration for 60 days,at the last day of performing intragastric administration, taking out the blood from eyeball, then took out the brains,thymus,spleens,livers and skin, and measure the viscera index of brains,thymus and spleens. Finally detected the superoxide dismutase( SOD),catalase( CAT),glutathione peroxidase( GSH-Px),malondialdehyde( MDA),total antioxidant capacity( T-AOC). Result: Compared with the blank group group,the model group the viscera index of brains,thymus and spleens decreased obviously( P〈0.05,P〈0.01),T-AOC,SOD,CAT in serum,livers,brains and skin decreased,and MDA increased. The viscera index of brains,thymus and spleens have increased a lot( P〈0.05,P〈0.01) after treateded EEFC. All dosage groups of EEFC could increase the activity of SOD,CAT in serum,livers,brains and thymus( P〈0.05,P〈0.01),and reduce the content of MDA in these organizations( P〈0.05, P〈0.01), meanwhile the T-AOC was increased obviously( P〈0.05,P〈0.01),and increase the activity of GSH-Px in serum and skin. Conclusion: EEFC has anti-aging effect on the subacute aging model mice which were Established by the injection of D-galactose.
