摘要
目的:研究组分配伍四逆汤对甲状腺功能减退症脑神经递质的影响。方法:将96只大鼠随机分为空白组、模型组、优甲乐阳性对照组(0.01 g·kg-1),组分配伍四逆汤高、中、低剂量组(9.6,4.8,2.4 g·kg-1)。除空白组外,其他组连续灌胃丙基硫氧嘧啶(PTU)溶液0.01 g·kg-115 d造甲减模型,用组分配伍四逆汤高、中、低剂量和优甲乐对此动物模型进行治疗,连续给药1个月,治疗同时隔日给予1%PTU维持甲减状态,实验结束后,取脑组织,做10%脑匀浆,用酶联免疫吸附法测去甲肾上腺素(NA),5-羟色胺(5-HT)和多巴胺(DA)浓度。结果:模型组与空白组相比,其NA浓度明显降低(P<0.05),5-HT浓度显著升高(P<0.01),DA浓度明显降低(P<0.05)。与模型组相比,四逆汤高、中剂量、阳性药组,NA浓度有显著升高(P<0.01);四逆汤低、中、高剂量组、阳性对照组,5-HT浓度明显降低(P<0.05,P<0.01);四逆汤高、中剂量、阳性药组,DA浓度显著升高(P<0.01)。结论:四逆汤对甲状腺功能减退症脑内神经递质有一定的调节作用。
Objective: To study the brain neurotransmitter influences of component comptibility of Sini Tang( CCSNT) to hypothyroidism. Method: The hypothyroidism mode was made by 1% propylthiouracil( PTU)solution 15 days,treat the animal which has hypothyroidism with CCSNT of high,middle,low dose( 9. 6,4. 8,2. 4 g·kg^-1,ig) and positive drugs Euthyrox 0. 01 g·kg^-1,ig,at the same time the mice should give the PTU( 0. 01 g·kg^-1,ig); after the end of the experiment,take the brain and make 10% brain homogenates,use the enzyme-linked immunosorband assay( ELISA) kit to measure the norepinephrine( NA),5-hydroxytryptaphane( 5-HT) and dopamine( DA) concentration. Result: Compared with the blank group,in the model group the concentration of NA and DA are decreased( P〈0.05),concentrations of 5-HT is significantly increased( P〈0.01). Compared with model group,the concentration of NA and DA of CCSNT high dose,middle dose and the Euthynx group,are significantly increased( P〈0.05); the 5-HT level of all the treatment group are decreased( P〈0.05). Conclusion: CCSNT has same effect of Sini Tang to neurotransmitters of hypothyroidism.
