HPLC法测定九味羌活丸中5-羟甲基糠醛的含量

Content Determination of 5-hydroxymethyl Furfural in Jiuwei Qianghuo Pill by HPLC

目的：建立测定九味羌活丸中5-羟甲基糠醛含量的方法。方法：采用高效液相色谱法。色谱柱为Phenomenex Luna C18,流动相为甲醇-0.1%磷酸溶液（3∶97,V/V）,流速为1 ml/min,柱温为25℃,进样量为5μl,检测波长为280nm。结果：5-羟甲基糠醛进样量在0.066 64-0.333 2μg范围内与峰面积积分值呈良好的线性关系（r=0.999 2）;精密度、稳定性、重复性试验的RSD≤1.15%;平均加样回收率为100.84%,RSD=4.06%（n=6）。结论：该方法简便、快速,重复性好,可用于九味羌活丸中5-羟甲基糠醛的含量测定。

OBJECTIVE：To establish the method for the content determination of 5-hydroxym ethyl furfural in Jiuwei qianghuo pill. METHODS：HPLC method was adopted. The determination was performed on Phenomenex Luna C18 column with mobile phase consisted of methanol-0.1% phosphoricl acid（3 ∶ 97,V/V）at the flow rate of 1 ml/min. The column temperature was set at25 ℃,and sample size was 5 μl. The detection wavelength was set at 280 nm. RESULTS：The linear range of 5-hydroxym ethyl furfural was 0.066 64-0.333 2 μg（r=0.999 2）with an average recovery of 100.84%（RSD=4.06%,n=6）;RSDs of precision,stability and reproducibility tests were all lower than 1.15%. CONCLUSIONS：The method is simple,rapid and reproducible for the content determination of 5-hydroxymethyl ethyl furfural in Jiuwei qianghuo pills.
